Would it be the same as your classic tower defense game but just reskined as pixel dungeon? If so I don't really see the point; there's already way too many games like that.
However perhaps the reverse of tower defense would work; effectivy, it would be the same as the current pixel dungeon, but with lots more enemies, infinite levels, and rebalenced so exploring depths is unimportant (not sure how that would work) and it's much easier to find the exit. So the aim would be to get to the deapest depth.

What for? Is there something in PD which makes you think "oh, that'd gonna look awesome if it were in TD"? And why tower defense and not, say, strategy or another CoD-clone?
